Week 11

November 8th, 2011
                On this day I inoculated my unknown onto a dish which antibiotics would be placed to see what the bacteria is sensitive to.  I placed them in the numbered areas as followed: Area 1 was penicillin, 2 was vancomycin, 3 was novabiocin, 4 was tetracycline, 5 was erythromycin, and 6 was chlorophenol.  I put it in the incubator to see where the bacteria grew.  I also inoculated four tubes with my unknown to see if it ferments or not.  The substances were nitrate, indole, citrate, and urea.  I also put these in the incubator.

                When I checked the tubes the next day, although the bacteria did grow in them, the only substance that gave a positive reaction was the urea, which turned pink.  We also tested nitrate further, using sulfanilic acid and dimethyl-alpha-naphthylamine to see if instead the nitrate can be converted to nitrite or nitrogen gas.  When it turned red only after the substances were added (as well as zinc), then it was clear that it did not use nitrate at all, giving a negative result.  I also did an oxidase test by acquiring bacteria with a cotton swab and pouring an oxidase reagent on it.  It turned yellow, indicating a negative reaction.   When I checked my antibiotic test, bacteria were only growing in a small corner, and because of how little really grew, it was determined that it was sensitive to all antibiotics in the dish.
